Neuronal Synaptic Activity

Neuronal synaptic activity is the communication that occurs when neurons transmit signals to one another or to target cells, forming the basis of nervous system function. An action potential reaching a presynaptic terminal triggers calcium influx and neurotransmitter release into the synaptic cleft; these molecules bind postsynaptic receptors to produce excitatory or inhibitory changes in membrane potential. The balance and timing of synaptic activity shape neural circuits involved in sensation, movement, learning, and memory. Measuring synaptic transmission and plasticity helps researchers investigate brain development, neurological and psychiatric disorders, and the effects of potential therapeutics.

This procedure performs long-lasting in vivo intracellular recordings from single neurons during physiologically relevant cerebral states and after complete abolition of ongoing electrical activities, resulting in an isoelectric brain state. The physiological constants of the animal are carefully monitored during the transition to the artificial comatose condition.

Vibrodissociation of Neurons from Rodent Brain Slices to Study Synaptic Transmission and Image Presynaptic Terminals

This report demonstrates a technique for mechanical isolation of individual viable neurons retaining attached presynaptic boutons. Vibrodissociated neurons have the advantages of rapid production, excellent pharmacological control and improved space-clamp without influence from neighboring cells. This method can be used for imaging of synaptic elements and patch-clamp recording.

The preparation of acute brain slices from isolated hippocampi, as well as the simultaneous electrophysiological recordings of astrocytes and neurons in stratum radiatum during stimulation of schaffer collaterals is described. The pharmacological isolation of astroglial potassium and glutamate transporter currents is demonstrated.
