Single Neuron Activation

Single neuron activation is the controlled excitation of an individual nerve cell to produce a measurable electrical response, enabling researchers to examine how neurons encode and transmit information. Activation occurs when membrane depolarization reaches threshold, opening voltage-gated ion channels and generating an action potential that propagates along the axon. In neuroscience, researchers can evoke or monitor this response using targeted electrical stimulation, intracellular methods, or light-sensitive approaches, depending on the experimental design. Studying single neuron activation helps link cellular excitability to synaptic communication, neural circuit function, sensory processing, and behavior, while supporting investigations of neurological disorders and emerging neurotechnology.

Optogenetic techniques have made it possible to study the contribution of specific neurons to behavior. We describe a method in larval zebrafish for activating single somatosensory neurons expressing a channelrhodopsin variant (ChEF) with a diode-pumped solid state (DPSS) laser and recording the elicited behaviors with a high-speed video camera.
