Transgenic Mouse Model

A transgenic mouse model is a laboratory mouse whose genome has been intentionally modified to introduce, remove, or alter a gene, enabling researchers to study gene function in a living organism. Genetic material is introduced into embryonic cells or early embryos, and successfully modified mice can transmit the engineered change to their offspring; tissue-specific promoters may restrict gene expression to selected cell types. In neuroscience, these models help link genes to neuronal development, synaptic signaling, behavior, and neurological disease. They also support testing of disease mechanisms, therapeutic targets, and genetic contributions to brain disorders under controlled experimental conditions.

The method described here allows for the visualization of IL-8 promoter-dependent inflammation activation in the lungs of mice through non-invasive bioluminescence imaging (BLI). The same animal can be subjected to BLI multiple times for up to two months from the time of delivery of the luciferase reporter construct.

The intensely studied nematode worm Caenorhabditis elegans can be transgenically engineered to express the human β-amyloid peptide (Aβ). Induced expression of Aβ in C. elegans muscle leads to a rapid, reproducible paralysis phenotype that can be used to monitor treatments that modulate Aβ toxicity.

2014

Activation of latent mutations with adenovirus-Cre into the mammary ductal system results in a clinically relevant metastatic breast cancer. Incorporation of a YFP promoter allows tracking of distal metastatic tumor cells. This model is useful to study latent metastasis, anti-tumor immunity, and for designing novel immunotherapies to treat breast cancer.
