Questo processo è altamente regolato, e solo un mRNA che esiste in una ribonucleoparticella, può essere trasportato dal nucleo al citoplasma da tradurre.Il controllo traslazionale, è un altro punto cruciale per la regolazione dell'espressione genica. La regolazione può essere specifica, dove sono coinvolti un individuo o un sottoinsieme di mRNA, o generale, dove la maggior parte dei trascritti di mRNA sono influenzati. Regulation of Expression Occurs at Multiple Steps

L'espressione genica può essere regolata in quasi ogni passaggio, dal gene alla proteina. La trascrizione è il passaggio più comunemente regolamentato. Ciò comporta il legame delle proteine a brevi sequenze regolatrici sul DNA. Questa associazione può promuovere o inibire la trascrizione di un gene associato alla rispettiva sequenza.

La trascrizione determina la generazione di un precursore (pre-mRNA) costituito sia da esoni che da introni, che necessita di ulteriore elaborazione prima di essere tradotto in una proteina. Ciò avviene attraverso lo splicing dell'mRNA, che comporta la rimozione delle regioni non codificanti e la fusione di quelle codificanti. L'elaborazione dell'mRNA può anche essere utilizzata come meccanismo di regolazione attraverso la variazione dei modelli di splicing come il salto di alcuni esoni, lo splicing alternativo e l'inclusione di introni.

Anche l’aggiunta di una coda poli-A all’estremità 3’ e di un cap 5’ per produrre l’mRNA maturo sono punti regolatori durante l’elaborazione dell’RNA. La regolazione avviene attraverso la variazione del segnale di poliadenilazione, che determina dove verrà aggiunta la coda poli-A all'mRNA. In alcuni casi, è presente più di un segnale poli-A all’estremità 3’, il che modificherà la lunghezza della regione 3’ non tradotta, ma il prodotto proteico finale sarà lo stesso. Tuttavia, la stabilità e il potenziale di traduzione delle varianti dell'mRNA possono differire, il che può alterare la quantità di proteine prodotte. In altri casi, un ulteriore segnale poli-A è presente sull'introne o sull'esone all'interno della sequenza genetica che può portare a variazioni nei siti di splicing per la poliadenilazione e provocare proteine diverse dallo stesso filamento di pre-mRNA. L'aggiunta dei 5 segnali 'cap, costituito da guanosina metilata, è regolata da due meccanismi. Uno prevede la regolazione delle metiltransferasi che aggiungono il gruppo metilico alla guanosina, e l'altro avviene attraverso la regolazione delle vie di segnalazione cellulare che portano alla metilazione.

Successivamente, l'mRNA maturo deve essere trasportato dal nucleo al citoplasma attraverso i complessi dei pori nucleari (NPC) per essere tradotto. Questo è regolato dall'mRNA fino alla formazione di un complesso, noto come ribonucleo particella, con le proteine leganti l'RNA. Gli NPC consentono solo agli mRNA presenti nel complesso di passare nel citoplasma. Una volta che un mRNA entra nel citoplasma per la traduzione, può essere preso di mira individualmente o come parte di un gruppo attraverso regolazioni specifiche, oppure può subire una regolazione comune con tutti gli altri mRNA nel citoplasma. Nella regolazione specifica, particolari elementi transattivi, come proteine e diversi tipi di RNA, regolano la trascrizione. Nella regolazione generale, le proteine coinvolte nel meccanismo di traduzione vengono attivate o inibite, il che a sua volta influenza la traduzione di tutte le trascrizioni. Il meccanismo di regolazione traslazionale più comune è la modifica del fattore di inizio della traduzione.

L'espressione genica può anche essere regolata attraverso modifiche post-traduzionali, dove una modifica reversibile catalizzata da un enzima può alterare la funzione di una proteina. Una modifica post-traduzionale comune è la fosforilazione, che viene effettuata da enzimi noti come chinasi. La defosforilazione delle proteine, invece, viene effettuata da proteine note come fosfatasi. La fosforilazione di una proteina può provocare la sua attivazione o disattivazione e alterarne la funzione.

Cells control gene expression at several points between DNA and protein. The most common control point is transcription. At this stage, proteins bind to short regulatory DNA sequences and can either increase or reduce transcription of the linked gene.

After transcription, a precursor mRNA is made. This pre-mRNA contains exons, which code for protein, and introns, which do not. It must be processed before translation, and splicing removes the introns and joins the exons. Cells can also regulate gene expression through changes in splicing patterns, including skipping certain exons, alternative splicing, and including introns.

RNA processing also includes adding a 3' poly-A tail and a 5' cap to form mature mRNA. The poly-A tail is added at a site marked by a polyadenylation signal. In some cases, more than one poly-A signal is present at the 3' end. This can change the length of the 3' untranslated region without changing the final protein, but the mRNA variants may differ in stability and translation potential. In other cases, an extra poly-A signal in an intron or exon can change splicing during polyadenylation and produce different proteins from the same pre-mRNA strand.

The 5' cap is a methylated guanosine, and its addition is also regulated. One control route acts through methyltransferases, the enzymes that add the methyl group. Another route works through cellular signaling pathways that lead to methylation. After processing, mature mRNA must leave the nucleus through nuclear pore complexes. It does this by forming a ribonucleoparticle with RNA-binding proteins, and only mRNAs in this complex can pass into the cytoplasm.

Once mRNA reaches the cytoplasm, translation can be regulated in specific ways or in a more general way. Specific regulation affects particular transcripts through trans-acting elements such as proteins and different types of RNA. General regulation affects all cytoplasmic mRNAs by turning translation machinery proteins on or off. The most common translational control step is modification of the translation initiation factor. Gene expression can also change after a protein is made through post-translational modifications. A common example is phosphorylation, which is carried out by kinases, while phosphatases remove the phosphate group through dephosphorylation. Phosphorylation can activate or deactivate a protein and change its function.
