DNA Isolation

Il DNA deve essere isolato dalle cellule e tagliato in posizioni precise per molte applicazioni, come la tecnologia del DNA ricombinante.
Sebbene vengano utilizzati diversi tipi di metodi di estrazione del DNA per diversi tipi di cellule, ci sono tre fasi standard: lisi cellulare, rimozione delle proteine e recupero del DNA.
All'interno delle cellule eucariotiche, il DNA è impacchettato all'interno del nucleo. Pertanto, sia la membrana cellulare che la membrana nucleare devono essere rotte per isolare il DNA.
Questo passaggio può essere eseguito meccanicamente scomponendo le cellule attraverso la macinazione o la sonicazione, o chimicamente utilizzando detergenti ed enzimi per dissolvere parti della membrana cellulare.
Una volta che il contenuto delle cellule viene rilasciato, i detriti vengono separati dai componenti solubili mediante centrifugazione. Il surnatante recuperato contiene acidi nucleici e proteine idrosolubili.

isolamento del dna

I protocolli di isolamento del DNA possono essere rapidi e diretti o complessi e richiedere molto tempo a seconda del tipo e della qualità del DNA richiesto per l'ulteriore elaborazione. Ad esempio, l'estrazione del DNA plasmidico è un po' più complicata dell'estrazione del DNA genomico a causa della necessità di un metodo di lisi appropriato per separare il DNA plasmidico dal gDNA durante l'isolamento. Tipi di metodi di estrazione del DNA genomico

Lo scopo principale dei metodi di estrazione del DNA genomico è separare il gDNA dalle proteine, dall'RNA e da altro contenuto cellulare. Comprende quattro passaggi fondamentali: 1. Distruzione della struttura cellulare meccanicamente o utilizzando sostanze chimiche per ottenere il lisato cellulare 2. Protezione del DNA dalla degradazione durante la lavorazione 3. Separazione del DNA solubile dai detriti cellulari 4. Eluizione del DNA purificato.

La maggior parte dei protocolli di isolamento del DNA genomico sono metodi di estrazione in soluzione o in fase solida. I metodi basati su soluzioni si basano su fasi di precipitazione e centrifugazione per separare il DNA da altro materiale cellulare, seguite dall'estrazione organica o "salatura" per separare il DNA solubile dalle proteine cellulari. La precipitazione finale del DNA viene effettuata utilizzando etanolo. Al contrario, i metodi di estrazione in fase solida utilizzano un supporto solido, come matrici di silice o cellulosa, per legare il DNA, seguito dal lavaggio e dall'eluizione del DNA dal supporto solido. Genomic DNA extraction methods separate DNA from proteins, RNA, and other cell material. The process usually follows four main steps. First, the cell structure is disrupted with mechanical or chemical methods to make a cell lysate. Then the DNA is protected from damage during processing. After that, the soluble DNA is separated from cell debris. Finally, the purified DNA is eluted, or washed out, for later use.

DNA isolation can follow two main approaches: solution-based methods or solid-phase extraction methods. Solution-based methods use precipitation and centrifugation to separate DNA from other cellular material. They may also use organic extraction or salting out to remove proteins from the soluble DNA. The final DNA is then precipitated with ethanol.

Solid-phase extraction methods use a solid support, such as silica or cellulose matrices, to bind the DNA. The sample is then washed so other cellular components are removed. Next, the DNA is eluted from the solid support. These methods may use centrifugation, vacuum, or magnetic steps to separate the bound DNA during the process.

The best DNA extraction method depends on the sample type, how many samples must be processed, and the next step for the DNA. Plasmid DNA extraction is usually more complex than genomic DNA extraction because the lysis method must separate plasmid DNA from genomic DNA during isolation. In some cases, such as long-range DNA sequencing, specific protocols are needed to obtain enough high-quality DNA.
