Restriction Enzymes

Il DNA isolato può essere tagliato in siti specifici da enzimi chiamati endonucleasi di restrizione.
Gli enzimi di restrizione sono isolati dai batteri. Riconoscono e scindono il DNA in corrispondenza di quattro-otto nucleotidi specifici in una sequenza palindrroma; una sequenza che si legge allo stesso modo nella direzione da 5' a 3' su un filamento e nella direzione da 3' a 5' sul filamento complementare.
Se l'endonucleasi taglia simmetricamente tra due basi opposte l'una all'altra su filamenti di DNA complementari, i frammenti di DNA risultanti hanno estremità smussate.
Se l'endonucleasi fa tagli sfalsati attraverso i due filamenti di DNA, lasciando ogni frammento con una sporgenza di basi spaiate, allora si dice che le estremità dei frammenti di DNA sono appiccicose.

enzimi di restrizione

Gli enzimi di restrizione sono enzimi batterici utilizzati per tagliare il DNA in modo sequenza-specifico. Per scindere il DNA, si legano a specifiche sequenze palindromiche chiamate siti di restrizione. Tali sequenze di DNA palindromiche o ripetizioni invertite si trovano comunemente in regioni di significato funzionale, come l'origine della replicazione, i siti degli operatori genici e le regioni contenenti segnali di terminazione della trascrizione.

I batteri ospiti proteggono il proprio DNA genomico da questi enzimi metilando questi siti. Alcuni batteri hanno enzimi che hanno sia la capacità di tagliare il DNA che di metilarlo con la stessa specificità di sequenza. EcoRI agisce come un enzima di restrizione quando è un dimero di subunità identiche. Come monomero, agisce come una metilasi. Altri batteri hanno due enzimi diversi per svolgere ciascuna funzione. Questa strategia di restrizione e modifica impedisce ai virus batterici di attaccare il genoma batterico.

Poiché specie batteriche diverse producono enzimi di restrizione diversi, ciascun enzima ha un sito di restrizione unico e prende il nome dal batterio di origine. Ad esempio, EcoRI è isolato dal ceppo E.coli RY13.

Restriction enzymes are bacterial proteins that cut DNA at specific sequence sites. They recognize palindromic DNA sequences, also called inverted repeats, and cut the DNA in a sequence-specific way. These sites are often found in regions with important functions, such as the origin of replication, gene operator sites, and transcription termination signals.

Bacteria protect their own DNA by methylating these same sites. Methylation means adding chemical marks that keep the restriction enzyme from cutting the host genome. In some bacteria, one enzyme can both cut DNA and add methyl groups when it has the same sequence specificity.

EcoRI is a well-known example. It acts as a restriction enzyme when it is a dimer made of identical subunits. When it is a monomer, it acts as a methylase. Other bacteria use two separate enzymes to perform cutting and methylation. This restriction and modification system helps protect bacterial DNA from viral attack.

Each restriction enzyme comes from a particular bacterial species and has its own DNA target site. The enzyme name reflects its source, such as EcoRI from the E. coli strain RY13. When DNA is cut with a specific restriction enzyme, the fragments have the same sequence at their 5' and 3' ends. If a plasmid DNA and an insert are cut with the same enzyme, their ends match and can be ligated easily. The fragments are then often checked on an agarose gel to see whether the DNA length matches the expected size.
