Maxam-Gilbert Sequencing

Nel metodo Maxam-Gilbert, o scissione chimica, il DNA stampo viene prima denaturato e radiomarcato all'estremità 5' con un fosforo 32. 
La reazione di sequenziamento viene eseguita in quattro diverse provette di reazione con quattro diverse sostanze chimiche con la capacità di modificare diversi nucleotidi nel DNA. 
Ad esempio, l'acido formico attaccherà solo le purine, l'adenina o la guanina, mentre l'idrazina attaccherà solo le pirimidine - citosina o timina. Allo stesso modo, sostanze chimiche come il dimetilsolfato attaccheranno solo un singolo nucleotide.
La piperidina viene quindi utilizzata per scindere il DNA alle basi modificate, generando frammenti di DNA radiomarcati di diverse lunghezze.
Le quattro reazioni vengono quindi eseguite parallelamente su un gel e visualizzate mediante autoradiografia. La sequenza del DNA stampo può quindi essere decifrata leggendo l'autoradiografia dal basso verso l'alto

Nello stesso anno della scoperta del metodo di sequenziamento Sanger, un altro gruppo di scienziati, Allan Maxam e Walter Gilbert, dimostrarono il loro metodo di scissione chimica per il sequenziamento del DNA. Il metodo Maxam-Gilbert si basa sull'utilizzo di diverse sostanze chimiche in grado di scindere la sequenza del DNA in siti specifici, sulla separazione dei frammenti di DNA risultanti di dimensioni variabili mediante elettroforesi e sulla decifrazione della sequenza di DNA dalle bande di gel risultanti.

Sfide del metodo Maxam-Gilbert

Il metodo Maxam-Gilbert può essere utilizzato solo per risolvere in genere fino a 400 bp in una singola analisi, un valore comparativamente inferiore rispetto ad altri metodi di sequenziamento tradizionali come il sequenziamento Sanger. Inoltre, la necessità di quantità relativamente elevate di DNA modello, unita all'uso di radioisotopi e sostanze chimiche pericolose, rende il metodo Maxam-Gilbert meno favorevole per l'uso di routine. Chemical cleavage DNA sequencing uses specific chemicals to cut DNA at known sites. Allan Maxam and Walter Gilbert described this method in the same year that the Sanger sequencing method was discovered. It reads the DNA sequence by breaking purified DNA into fragments and then studying the fragment pattern.

The method depends on chemicals that cleave DNA at specific bases. The resulting fragments are separated by electrophoresis, which is a way to sort DNA pieces by size using an electric field. After that, the sequence is determined from the band pattern seen on the gel.

The Maxam-Gilbert method has important limits. It usually resolves up to about 400 base pairs in one run, which is less than some traditional sequencing methods such as Sanger sequencing. It also needs relatively large amounts of template DNA, and it uses radioisotopes and hazardous chemicals, so it is not ideal for routine use.

Even with these drawbacks, the method has one advantage. Because it works directly with purified DNA, it can be more accurate than Sanger sequencing. Today, it is used less often, but it still has value in some DNA fingerprinting methods and in DNA structural studies.

These technologies may be best investigated along with the standards from LS1 From Molecules to Organisms: Structure and Functions, as the students are learning about DNA and protein. Comparative DNA and protein sequence analysis is also a large part of current phylogenic research and could accompany LS4 Biological Change: Unity and Diversity. Students may also consider the pros and cons of biotechnical applications. Investigation of techniques utilized could come from field trips to witness the use of these techniques in practice, lab exercises, virtual labs, simulation activities, interviews with professionals, etc. Molecular techniques could include: PCR, electrophoresis, restriction enzyme digestion of DNA, DNA sequencing, plasmid-based transformation, transfection, etc. These techniques are used in fields of medicine, agriculture, biomedical engineering, forensic science, etc.
