L'immunocitochimica o ICC e l'immunoistochimica o IHC sono tecniche di colorazione che utilizzano anticorpi legati agli enzimi per visualizzare e quantificare proteine o antigeni specifici in cellule in coltura o sezioni di tessuto.
Nell'ICC, un singolo strato di cellule bersaglio viene coltivato su un vetrino coprioggetti. Le cellule vengono fissate con un agente reticolante per prevenire la degradazione enzimatica dell'antigene, quindi trattate con un detergente per rendere permeabile la membrana cellulare.
Nell'IHC l'intero tessuto è fissato e incorporato in un blocco di paraffina. Quindi, il campione viene sezionato in fette sottili, che possono essere posizionate su vetrini. Infine, la paraffina viene rimossa per recuperare l'antigene.
Una volta che gli antigeni sono accessibili nel campione, vengono legati da anticorpi primari specifici per il bersaglio legati a enzimi coniugati come la perossidasi di rafano.
L'enzima catalizza l'ossidazione della colorazione DAB in un precipitato marrone consentendo la visualizzazione della proteina di interesse.
Se il segnale rilevato è debole, vengono utilizzati anticorpi secondari con enzimi coniugati per legare un anticorpo primario non marcato sulla proteina bersaglio per un'ulteriore amplificazione del segnale.

Immunocytochemistry and Immunohistochemistry

L'immunocitochimica (ICC) e l'immunoistochimica (IHC) sono tecniche che utilizzano anticorpi per verificare la presenza di proteine o antigeni specifici in un campione. La tecnica fu pubblicata per la prima volta da Albert Coons nel 1941 per rilevare la presenza di antigene pneumococcico in sezioni di tessuto di topi infetti da Pneumococco. L'immunocitochimica aiuta la localizzazione di proteine o antigeni in singole cellule come cellule del sangue, cellule staminali, ecc., mentre l'immunoistochimica fa lo stesso per i campioni di tessuto.

Queste tecniche possono essere colorimetriche se vengono utilizzati anticorpi coniugati con enzimi o se gli anticorpi marcati con fluoroforo basati su fluorescenza localizzano le proteine o gli antigeni di interesse. Il prodotto colorato ottenuto tramite la reazione enzimatica o la fluorescenza emessa dal fluoroforo viene quindi visualizzato al microscopio ottico o ad immunofluorescenza.

Queste tecniche possono utilizzare anticorpi primari o secondari contrassegnati con un enzima o un fluoroforo. Se viene utilizzato un anticorpo primario, la tecnica viene definita ICC diretta o IHC, poiché gli anticorpi coniugati si legano direttamente all'epitopo della proteina o dell'antigene di interesse. In modo indiretto, un anticorpo secondario marcato con l'enzima o il fluoroforo si lega all'anticorpo primario attaccato all'epitopo della proteina o dell'antigene di interesse. Il segnale emesso al momento del legame di un anticorpo secondario è più forte del segnale di un anticorpo primario.

Entrambe le tecniche variano leggermente nelle fasi di preparazione del campione. Nell'ICC viene generato uno strato di cellule in monocoltura, che vengono poi trattate con agenti fissanti come la formaldeide per prevenire la degradazione enzimatica degli antigeni, seguito da un trattamento con detergenti come Triton X o Tween 20 per rendere la membrana cellulare permeabile all'ingresso di anticorpi. Allo stesso modo, nell'IHC, i campioni di tessuto vengono prima trattati con una soluzione di fissaggio, seguita dall'inclusione in cera di paraffina per prevenire danni ai tessuti fragili. I campioni di tessuto vengono quindi sezionati in fette sottili e trattati con detergente prima dell'introduzione degli anticorpi.

Una volta che le cellule o i vetrini di tessuto sono stati trattati con detergente, vengono aggiunti gli anticorpi. Nell'ICC o IHC diretto, viene aggiunto l'anticorpo primario marcato con fluoroforo o enzimi e, dopo l'incubazione, vengono visualizzati sotto immunofluorescenza o microscopio ottico. Nell'ICC o IHC indiretto, i campioni vengono prima incubati con l'anticorpo primario, seguito da un lavaggio per rimuovere gli anticorpi non legati. Viene quindi introdotto l'anticorpo secondario, che si lega all'anticorpo primario ed emette fluorescenza o prodotto colorato dopo l'aggiunta del colorante. I campioni sono ora pronti per la visualizzazione.

Immunocytochemistry (ICC) and immunohistochemistry (IHC) use antibodies to detect specific proteins or antigens in samples. ICC focuses on individual cells, such as blood cells and stem cells. IHC is used for tissue samples. These methods were first published by Albert Coons in 1941, when they were used to detect pneumococcal antigen in mouse tissue sections from mice infected with Pneumococcus.

Both techniques can produce either a colored signal or a fluorescent signal. Enzyme-conjugated antibodies create a colorimetric result through a chemical reaction. Fluorophore-tagged antibodies produce fluorescence that can be seen with an immunofluorescence microscope. The signal can come from a primary antibody or from a secondary antibody, depending on the method used.

In direct ICC or IHC, the primary antibody is tagged with an enzyme or fluorophore and binds directly to the epitope, or target site, on the protein or antigen. In indirect ICC or IHC, an unlabeled primary antibody binds first, and then a tagged secondary antibody binds to it. The indirect method usually gives a stronger signal than the direct method.

The sample preparation steps are slightly different for cells and tissues. In ICC, a monoculture layer of cells is first fixed with a solution such as formaldehyde to prevent antigen breakdown. The cells are then treated with detergents like Triton X or Tween 20 so antibodies can enter the cells. In IHC, tissue samples are first fixed, then embedded in paraffin wax to protect fragile tissue. The tissue is cut into thin slices and treated with detergent before antibodies are added.

After detergent treatment, the antibodies are introduced. In direct ICC or IHC, the tagged primary antibody is added and the sample is later viewed under an optical or immunofluorescence microscope. In indirect ICC or IHC, the sample is first incubated with the primary antibody and washed to remove any unbound antibodies. The secondary antibody is then added, and it produces fluorescence or a colored product after dye addition. The slide is then ready for visualization.
