La microscopia crioelettronica o crio-EM esamina campioni biologici congelati a temperature criogeniche inferiori a -150 °C.
A differenza dell'EM convenzionale, la cryo-EM non richiede il fissaggio e la disidratazione di un campione, il che aiuta a preservare lo stato nativo delle strutture biologiche.
Gli elettroni ad alta energia utilizzati nell'EM possono danneggiare i legami chimici di un campione. Il congelamento del campione aiuta a prevenire questo danno, consentendo di ottenere immagini con risoluzione quasi atomica.
Nella crio-EM, piccoli campioni biologici in soluzione vengono vitrificati, cioè congelati rapidamente, in etano liquido. La vitrificazione fissa il campione nel suo stato nativo in un sottile strato d'acqua e l'acqua nel campione forma uno stato simile al vetro senza la comparsa di cristalli di ghiaccio.
I campioni più spessi, come cellule e tessuti, non possono essere vetrificati in un sottile strato liquido. Pertanto, questi campioni possono essere studiati utilizzando il metodo crio-EM delle sezioni vitreali. In questo metodo, un campione viene prima vetrificato mediante congelamento ad alta pressione e poi tagliato in sezioni ultrasottili a -140 °C prima di essere osservato utilizzando la crio-EM.

Cryo-electron Microscopy

La microscopia elettronica convenzionale (EM) comporta la disidratazione, la fissazione e la colorazione dei campioni biologici, che distorcono lo stato nativo delle molecole biologiche e provocano numerosi artefatti. Inoltre, il fascio di elettroni ad alta energia danneggia il campione e rende difficile ottenere immagini ad alta risoluzione. Questi problemi possono essere risolti utilizzando la crio-EM, che utilizza campioni congelati e fasci di elettroni più delicati. La tecnica è stata sviluppata da Jacques Dubochet, Joachim Frank e Richard Henderson, per la quale hanno vinto il Premio Nobel per la Chimica nel 2017.

Le tecniche di diffrazione dei raggi X e di risonanza magnetica nucleare (NMR) vengono utilizzate anche per ottenere strutture ad alta risoluzione di biomolecole o studiarne i cambiamenti conformazionali. Tuttavia, per ottenere la struttura a raggi X, una molecola deve essere cristallizzata, cosa che potrebbe non essere possibile ogni volta. Anche se una molecola è cristallizzabile, il processo di cristallizzazione può alterare la sua struttura biomolecolare, che non è più rappresentativa della struttura nativa. Cryo-EM non richiede un campione cristallizzato e consente inoltre la visualizzazione dei movimenti molecolari o delle interazioni delle biomolecole nel loro stato nativo. Allo stesso modo, la tecnica NMR è limitata solo a proteine solubili relativamente piccole e non è adatta per le proteine non polari incorporate nelle membrane cellulari. D'altra parte, la crio-EM consente lo studio di proteine più grandi, recettori legati alla membrana o complessi biomolecolari.

Cryo-EM è ampiamente utilizzato in biochimica per studiare la struttura, la funzione e l'interazione delle biomolecole. Gli agenti infettivi, come i virus, possono essere identificati e studiati utilizzando la crio-EM. Cryo-EM, or cryo-electron microscopy, lets scientists image biomolecules in their frozen, native state. It avoids the dehydration, fixation, and staining used in conventional electron microscopy, which can distort samples and create artifacts. It also reduces damage from the electron beam by using gentler imaging conditions.

This method is useful when other structure tools have limits. X-ray diffraction needs a molecule to form a crystal, and that is not always possible. Even when crystallization works, it can change the biomolecule’s structure so it no longer matches the native form.

Cryo-EM also fills gaps left by nuclear magnetic resonance, or NMR. NMR is mainly used for relatively small soluble proteins and is not suited to non-polar proteins in cell membranes. Cryo-EM can be used to study larger proteins, membrane-bound receptors, and biomolecular complexes.

Researchers use cryo-EM to study biomolecule structure, function, and interactions. It can also help identify and analyze infectious agents such as viruses. During the Zika virus outbreak in Brazil, cryo-EM was used to build a 3D image of the virus so potential antiviral drug targets could be found. The technique was developed by Jacques Dubochet, Joachim Frank, and Richard Henderson, who shared the 2017 Nobel Prize in Chemistry.
