High Temperature Preparation

High temperature preparation is a biochemistry approach that uses controlled heating to condition, extract, or stabilize biological samples before analysis. Elevated temperatures can accelerate solubilization, disrupt molecular interactions, denature proteins, and reduce enzymatic activity, while the specific temperature and exposure time determine which components remain intact. Researchers apply this strategy to prepare lysates, release biomolecules from complex materials, and improve the consistency of downstream assays such as electrophoresis, chromatography, and nucleic acid analysis. Because excessive heat can cause degradation or unwanted modification, careful control of heating conditions is essential for reliable biochemical measurements.

Slippery surfaces provide a new way to solve the adhesion problem. This protocol describes how to fabricate slippery surfaces at high temperatures. The results demonstrate that the slippery surfaces showed anti-wetting for liquids and a remarkable anti-adhesion effect on soft tissues at high temperatures.

The laser heated diamond anvil cell combined with synchrotron micro-diffraction techniques allows researchers to explore the nature and properties of new phases of matter at extreme pressure and temperature (PT) conditions. Heterogeneous samples can be characterized in situ under high pressure by 2D mapping and combined powder, single-crystal and multigrain diffraction approaches.
