Adipose-derived Stem Cells

Adipose-derived stem cells (ADSCs) are multipotent mesenchymal stromal cells obtained from adipose tissue, providing an accessible cell source for tissue engineering and regenerative research. They are typically isolated from the stromal vascular fraction after enzymatic digestion and expanded in culture; under defined biochemical and mechanical cues, they can differentiate toward adipogenic, osteogenic, and chondrogenic lineages while releasing paracrine signals. In bioengineering, researchers combine ADSCs with biomaterial scaffolds, hydrogels, or engineered constructs to support tissue formation, repair, and vascularization. Their relative abundance and minimally invasive harvest make them useful for studying cell-material interactions and developing personalized approaches to regenerative medicine.

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The multipotent pADSC are used to delineate processes of adipocyte differentiation and study transdifferentiation into multiple cell lineages of mesodermal mesenchyme or further lineages of ectoderm and endoderm for regenerative studies.

Isolating Adipose-Derived Stem Cells from Murine Periaortic Adipose Tissue

2025

This video demonstrates a technique for isolating and culturing fluorescently labeled neural crest-derived adipose-derived stem cells (ADSCs) from murine periaortic arch adipose tissue. Following isolation, the NCADSCs are detected, sorted using fluorescence-activated cell sorting, and subsequently cultured on a plate, promoting cell proliferation.

We present the techniques required to isolate the stromal vascular fraction (SVF) from mouse inguinal (subcutaneous) and perigonadal (visceral) adipose tissue depots to assess their gene expression and collagenolytic activity. This method includes the enrichment of Sca1high adipose-derived stem cells (ASCs) using immunomagnetic cell separation.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
