Airway Wall Model

An airway wall model is an engineered or reconstructed system that reproduces key structural and functional features of the airway wall for studying respiratory biology. Bioengineers combine airway epithelial cells, smooth muscle cells, extracellular matrix, and biomaterial scaffolds to recreate the wall’s layered organization, then apply relevant chemical or mechanical conditions to assess barrier function, inflammation, contraction, and tissue remodeling. These models support research on asthma, chronic obstructive pulmonary disease, infection, and airway injury while providing platforms for drug testing and therapeutic development. By complementing animal studies, airway wall models can help clarify disease mechanisms and improve the design of patient-relevant treatments.

Advancements in biomaterial technologies enable the development of three-dimensional multi-cell-type constructs. We have developed electrospinning protocols to produce three individual scaffolds to culture the main structural cells of the airway to provide a 3D in vitro model of the airway bronchiole wall.

This paper describes different methods of growing Pseudomonas aeruginosa biofilms on cultured human airway epithelial cells. These protocols can be adapted to study different aspects of biofilm formation, including visualization of the biofilm, staining of the biofilm, measuring the colony forming units (CFU) of the biofilm, and studying biofilm cytotoxicity.

A technique called Comprehensive Microarray Polymer Profiling (CoMPP) for the characterisation of plant cell wall glycans is described. This method combines the specificity of monoclonal antibodies directed to defined glycan-epitopes with a miniature microarray analytical platform allowing screening of glycan occurrence in a broad range of biological contexts.
