Cell Spheroid Formation

Cell spheroid formation is the process of assembling living cells into compact, three-dimensional aggregates that better represent the organization of many tissues than flat cell cultures. In bioengineering, cells are encouraged to cluster under nonadherent or confined conditions, where cell–cell adhesion, extracellular matrix production, and compaction drive aggregate formation and stabilization. Spheroids can reproduce gradients of oxygen, nutrients, and signaling molecules that arise within larger cellular structures, creating useful models of tissue behavior. They support tissue engineering, regenerative research, disease modeling, and drug screening by enabling researchers to study cell interactions, viability, and responses in a more physiologically relevant three-dimensional environment.

The uneven loss of medium from microtiter plates affects the reproducibility of uniform multicellular tumor spheroid formation. Improving culture conditions to reduce significant medium loss will improve the reproducibility of spheroid formation and the results of spheroid-based assays using the liquid-overlay technique.

Here, we describe a rapid and flexible protocol for the formation of 3D cell spheroids through cell aggregation. This is easily adapted to multiple cell types and is suitable for use in a variety of applications including cell migration, invasion, or anoikis assays, and for imaging and quantifying cell-matrix interactions.

This video demonstrates a method to establish a three-dimensional tumor spheroid and T cell co-culture. An agarose cast with microwells containing tumor spheroids is prepared, followed by the addition of T cells. After incubation, this setup allows studying the interaction of T cells with tumor spheroids in a controlled three-dimensional environment.
