Insulin Producing Cells

Insulin-producing cells are specialized pancreatic cells, primarily beta cells, that regulate blood glucose by synthesizing and secreting insulin, making them central to metabolic control and diabetes research. In response to elevated glucose, beta cells metabolize the sugar, increase ATP production, close ATP-sensitive potassium channels, and trigger calcium-dependent insulin release from secretory granules. Bioengineering approaches seek to generate, maintain, or assemble these cells using stem cell differentiation, biomaterials, tissue engineering, and encapsulation strategies. These systems support studies of diabetes mechanisms, drug screening, and the development of cell-based therapies designed to restore glucose-responsive insulin production while improving graft survival and function.

Endothelial Cell Co-culture Mediates Maturation of Human Embryonic Stem Cell to Pancreatic Insulin Producing Cells in a Directed Differentiation Approach

The current study describes a directed differentiation approach in inducing pancreatic differentiation of human embryonic stem cells. Of great significance is the finding that endothelial cell co-culture mediates maturation of human embryonic stem cell derived pancreatic progenitors into insulin expressing cells.

The differentiation of stem cells into islet cells provides an alternative solution to conventional diabetes treatment and disease modeling. We describe a detailed stem cell culture protocol that combines a commercial differentiation kit with a previously validated method to aid in producing insulin-secreting, stem cell-derived islets in a dish.

In this protocol, we provide detailed steps for the isolation and characterization of rat epididymal Ad-MSCs, followed by a simple, short protocol for the generation of IPCs from the same rat Ad-MSCs.

Conserved insulin signaling pathways found in the fruit fly Drosophila melanogaster make this organism a potential tool for modeling metabolic disorders including type II diabetes. Epigenetic Conversion as a Safe and Simple Method to Obtain Insulin-secreting Cells from Adult Skin Fibroblasts

Here, a new method that allows the conversion of adult skin fibroblasts into insulin-secreting cells is presented. This technique is based on epigenetic conversion, does not involve the use of retroviral vectors nor the acquisition of a stable pluripotent state. It is therefore highly promising for translational medicine applications.
