K-glutamate Calibration

K-glutamate calibration is the quantitative standardization of measurements for potassium glutamate, an ionic metabolite and commonly used component of biochemical systems. It works by measuring reference solutions with known K-glutamate concentrations, relating the resulting instrument signal to concentration, and using that calibration relationship to determine values in unknown samples under matched conditions. In bioengineering, reliable calibration supports controlled preparation of cell-free reaction mixtures, media, and experimental buffers, where glutamate concentration and ionic balance can influence biochemical activity. Consistent measurements improve reproducibility, enable comparison across experiments, and help researchers validate sensors, analytical workflows, and engineered biological systems.

Generally, a set of standard samples are made at various concentrations with a range than includes the unknown of interest and the instrumental response at each concentration is recorded. For more accuracy and to understand the error, the response at each concentration can be repeated so an error...

This video demonstrates evaluating glutamate release and clearance at single corticostriatal synapses in a mouse brain slice. Electrical stimulation facilitates glutamate release at the synapse. The released glutamate alters sensor protein's fluorescence, which decays over time, indicating release and clearance efficiency.

In Vivo Real-Time Glutamate Monitoring Using an Enzyme-Coated Microelectrode Array

In this video, a Multielectrode Array (MEA) coated with glutamate Oxidase is implanted into the brain tissue of an anesthetized mouse. Glutamate solution is injected into the extracellular space where the MEA detects the glutamate levels, enabling real-time monitoring of glutamate uptake by the astrocytes.
