Tetrahydrofuran Leaching

Tetrahydrofuran leaching is a solvent-based separation process that removes soluble components from a solid or porous material using tetrahydrofuran (THF), an organic solvent. THF penetrates the material, dissolves compatible polymers, additives, or residual compounds, and transports them outward through diffusion; subsequent rinsing, drying, or solvent evaporation leaves a modified matrix. In bioengineering, this approach can help process polymeric biomaterials, isolate material fractions, and adjust surface composition or internal porosity before biological testing. Because leaching efficiency depends on solvent compatibility, contact time, temperature, and material structure, the method also supports controlled fabrication and characterization of engineered biological interfaces.

We present an assay for easy quantification of metals introduced to samples prepared using immobilized metal affinity chromatography. The method uses hydroxynaphthol blue as the colorimetric metal indicator and a UV-Vis spectrophotometer as the detector.
