Vaccinia Topoisomerase

Vaccinia topoisomerase is a poxvirus-encoded type I DNA topoisomerase that regulates DNA topology by relieving torsional stress during genome replication and other DNA transactions. The enzyme recognizes specific DNA sequences, cleaves one strand through a transient covalent bond between an active-site tyrosine and the DNA phosphate, allows controlled rotation, and then religates the strand. In bioengineering, this well-characterized reaction supports studies of DNA topology, enzyme mechanism, and sequence-specific cleavage. Engineered variants and biochemical assays can also aid protein design, molecular diagnostics, and screening for compounds that disrupt topoisomerase activity.

We describe the usage of a fluorescent reporter vaccinia virus that enables real-time measurement of viral infectivity and gene expression through the stage-specific expression of spectrally distinct reporter fluorophores. We detail a plate-based method for accurately identifying the stage at which virus replication is affected in response to small molecule inhibition.
