Hepatitis B Virus

Hepatitis B virus (HBV) is a DNA virus that infects liver cells and can cause acute or chronic hepatitis, making it an important subject in biology and public health. After entering hepatocytes, HBV delivers partially double-stranded DNA to the nucleus, where it is repaired into covalently closed circular DNA that directs viral RNA and protein production; reverse transcription then converts pregenomic RNA into new viral DNA within assembling particles. Persistent infection can lead to cirrhosis and hepatocellular carcinoma, while immune responses contribute to liver injury. Studying HBV supports vaccine development, antiviral therapy, diagnostic testing, and research into viral persistence and liver disease.

A Protocol for Analyzing Hepatitis C Virus Replication

Cited by 2 •

2014

Hepatitis C Virus (HCV) is a major human pathogen that causes liver disorders, including cirrhosis and cancer. An HCV infectious cell culture system is essential for understanding the molecular mechanism of HCV replication and developing new therapeutic approaches. Here we describe a protocol to investigate various stages of the HCV replication cycle.

Here we describe a newly developed hepatitis B virus (HBV) reporter system to monitor the early stages of the HBV life cycle. This simplified in vitro system will aid in the screening of anti-HBV agents using a high-throughput strategy.

Targeting Liver Tumors with Oncolytic Viruses via the Hepatic Artery

2025

The video demonstrates locoregional delivery of oncolytic viruses through hepatic arterial injection as a therapeutic approach for hepatocellular carcinoma. These viruses selectively replicate in tumor cells, inducing cell lysis and releasing tumor cell-associated antigens. This process triggers immunogenic cell death, inducing an anti-tumor response.

(2014)This video demonstrates the fluorescence focus assay used to quantify infectious Hepatitis C virus in cell culture. It demonstrates the serial dilution of viral supernatant, the infection of hepatoma cells, and the detection of viral replication sites using antibody-based fluorescence microscopy.

Harvesting Hepatitis E Virus Particles from Human Liver Cancer Cells

Source: Meister, T. L., et al. A Cell Culture Model for Producing High Titer Hepatitis E Virus Stocks. J. Vis. Exp. (2020).This video demonstrates the process of isolating hepatitis E virus (HEV) particles from transfected human hepatocellular carcinoma cells. Extracellular viral particles are collected from filtered supernatants, while intracellular particles are released by subjecting the cells to repeated freeze-thaw cycles. The lysate is centrifuged, and the supernatant containing HEV...
