Nematode Microbiota

Nematode microbiota are the communities of microorganisms that live in or on nematodes, including bacteria, fungi, and other microbes, and they can influence host biology and ecological function. Microbes colonize nematode surfaces or intestinal environments, where interactions involving nutrient exchange, competition, chemical signaling, and host immune responses shape community composition and persistence. Studying these associations helps explain nematode development, metabolism, behavior, and adaptation to environmental conditions. In biology and agriculture, nematode microbiota research supports investigations of host–microbe relationships, plant disease, soil ecosystems, and biological control, while also identifying microbial functions relevant to biotechnology and parasite management.

Caenorhabditis elegans is one of the main model species in biology, yet almost all research is performed in the absence of its naturally associated microbes. The methods described here will help to improve our understanding of the diversity of associated microbes as a basis for future functional C. elegans research.

Visualizing Bacteria in Nematodes using Fluorescent Microscopy

To study the mutualism between Xenorhabdus bacteria and Steinernema nematodes, methods were developed to monitor bacterial presence and location within nematodes. The experimental approach, which can be applied to other systems, entails engineering bacteria to express the green fluorescent protein and visualizing, using fluorescence microscopy bacteria within the transparent nematode.

An In Vivo Assay to Detect Mitophagy in Transgenic Nematodes

This video demonstrates an in vivo assay to study mitophagy in Caenorhabditis elegans. Upon exposing the nematodes to a mitophagy-inducing drug, the occurrence of mitophagy is assessed by co-localization between DCT-1, a mitophagy receptor on the damaged mitochondria, and the autophagosomal membrane protein LGG-1.

Here we present several protocols for quantifying predatory feeding behavior found in the model nematode Pristionchus pacificus and its relatives. Additionally, we provide methods for analyzing predatory feeding adaptations including mouth structures and teeth.
