Lysine Discharge Assay

The lysine discharge assay is a biochemical method that measures the release of lysine from lysyl-tRNA, helping characterize reactions that remove amino acids from charged transfer RNA. In the assay, lysyl-tRNA is incubated with an enzyme or protein under defined conditions, and deacylation is quantified by detecting the lysine released or the remaining aminoacyl-tRNA. This approach is used to investigate tRNA quality control, aminoacyl-tRNA synthetase editing, and other mechanisms that protect translational accuracy. By measuring discharge rates and substrate specificity, researchers can compare enzymatic activity, examine catalytic defects, and clarify how cells prevent incorrectly charged tRNAs from entering protein synthesis.
