Sac Dissection

Sac dissection is a biological technique for opening and examining a sac-like anatomical structure while preserving its walls, contents, and surrounding tissues. The procedure typically involves exposing the sac, separating it from adjacent structures, and making a controlled incision with fine instruments, often under a stereomicroscope, to prevent damage and permit detailed observation. Researchers and students use sac dissection to study anatomy, tissue organization, developmental changes, and the contents or function of specialized compartments. By linking visible structure with biological role, this method supports comparative anatomy, developmental biology, physiology, and laboratory-based investigation of normal or altered specimens.

Dissection of the Endolymphatic Sac from Mice

This study describes how to dissect the endolymphatic sac from the inner ear of the mouse at different ages. The result of a similar dissection is shown in an Slc26a4-null mouse model of enlargement of the endolymphatic sac. A transgenic mouse with a fluorescent reporter expressed in the endolymphatic sac is presented as a model to readily visualize the endolymphatic sac, and educational tool.
