Microrna Target Validation

MicroRNA target validation is the experimental process of confirming whether a microRNA directly regulates a specific messenger RNA, an essential step in linking post-transcriptional control to cellular function. Researchers typically test the predicted microRNA binding site by placing the target sequence, often from an mRNA 3′ untranslated region, downstream of a reporter gene and measuring changes in reporter activity after microRNA introduction or inhibition. Mutating the binding site can determine whether regulation is sequence-specific, while quantitative PCR and protein analyses assess effects on endogenous transcripts and translation. These approaches clarify gene regulatory networks involved in development, disease, and therapeutic research.

MicroRNAs (miRNAs) are important regulators of gene expression and have been shown to play a role in numerous biological processes. To better understand miRNA-UTR interactions, we have created a genome-wide collection of 3 UTR luciferase reporters paired with a novel luciferase gene and assay reagent, the LightSwitch system.
