Cuticle Visualization

Cuticle visualization is the use of imaging and contrast-enhancement methods to reveal the structure, distribution, and surface properties of a cuticle, a protective outer layer found in plants and invertebrates. Depending on the specimen, microscopy can highlight cuticular boundaries, surface patterns, and associated structures by exploiting differences in optical properties or by applying stains or fluorescent labels that bind to particular components. In biology, these observations help researchers compare development, assess barriers to water loss or environmental stress, characterize insect morphology, and connect cuticle organization with functions such as protection, growth, and interaction with the environment.

Aerial plant organs are protected by the cuticle, a supramolecular biopolyester-wax assembly. We present protocols to monitor selective removal of epi- and intracuticular waxes from tomato fruit cuticles on molecular and micro scales by solid-state NMR and atomic force microscopy, respectively, and to assess the cross-linking capacity of engineered cuticular biopolyesters.

Lipid polyesters constitute the structural components of two cell wall modifications, the plant cuticle and suberin-containing diffusion barriers. In this video, we describe a method to depolymerize cutin from whole delipidated leaves. The method can be applied to investigating mutants compromised in either cutin or suberin biosynthesis.
