Middlebrook 7h10

Middlebrook 7H10 is a selective, solid culture medium used to grow and isolate mycobacteria, including organisms responsible for tuberculosis, under laboratory conditions. Its mineral-salt base supplies essential nutrients, while glycerol and OADC enrichment provide carbon, proteins, vitamins, and other growth factors; malachite green suppresses many contaminating microorganisms, and agar creates a surface for colony development. In biology and clinical microbiology, Middlebrook 7H10 supports the recovery and characterization of slow-growing mycobacteria, colony counting, and agar-based drug susceptibility testing. Its defined composition also helps researchers compare growth patterns and investigate antimicrobial responses reproducibly.

Source: Bernut, A., et al., Deciphering and Imaging Pathogenesis and Cording of Mycobacterium abscessus in Zebrafish Embryos. J. Vis. Exp. (2015).This video demonstrates a protocol for isolating and quantifying Mycobacterium abscessus from an infected zebrafish embryo. The procedure involves embryo euthanasia, tissue lysis, and homogenization to release the bacteria, followed by centrifugation and resuspension in a surfactant buffer. Finally, the bacterial suspension is serially diluted, plated...
