Natural Killer Cells

Natural killer (NK) cells are innate immune lymphocytes that identify and eliminate infected, stressed, or transformed cells without prior antigen-specific sensitization. They integrate signals from activating and inhibitory surface receptors, then release perforin and granzymes to trigger target-cell death or produce cytokines such as interferon-gamma to coordinate broader immune responses. In biology and biomedical research, NK cells provide a model for studying immune surveillance, host-pathogen interactions, and tumor immunity. Their ability to recognize altered cells rapidly supports investigations into cancer immunotherapy, antiviral defenses, transplantation, and strategies that enhance targeted cell killing.

Ex Vivo Expansion of Natural Killer Cells from Peripheral Blood Mononuclear Cells

This video demonstrates a technique for the expansion of natural killer (NK) cells directly from peripheral blood mononuclear cells (PBMCs) using a feeder cell line. PBMCs are co-cultured with genetically modified lymphoblastoid feeder cells expressing membrane-bound interleukin-21 (mIL-21), and the culture is supplemented with the cytokines interleukin-2 (IL-2) and interleukin-15 (IL-15). Together, this cocktail of ligands leads to the prolonged expansion of NK cells with high purity.

Isolating Invariant Natural Killer T Cells from a Mouse Model

This video illustrates a method for isolating invariant natural killer T cells, also known as iNKT cells. The procedure begins with injecting a synthetic glycolipid into a mouse to stimulate the activation and proliferation of iNKT cells in the spleen. Subsequently, single-cell suspensions are prepared from the spleen and incubated with iNKT-cell-recognizing complexes labeled with fluorophores, followed by magnetic bead-based separation, to isolate the iNKT cells.
