Contralateral Gland Analysis

Contralateral Gland Analysis is a comparative method that examines the gland opposite a tumor-bearing, diseased, or treated gland to clarify cancer-related changes. By evaluating paired tissues under comparable biological conditions, researchers can compare morphology, cellular composition, molecular markers, or gene expression and distinguish local effects from changes shared across the organism. In cancer research, this approach can provide an internal reference for assessing tumor-associated alterations, treatment responses, and disease progression. It is especially useful in preclinical studies, where paired-gland comparisons can reduce variability and strengthen interpretation of experimental findings.

We developed a method to successfully remove, process, section, and stain, for histopathological evaluation, mammary tissue that had originally been fixed on slides as whole mounts. This method may promote the collection and evaluation of mammary gland whole mounts in reproductive and developmental test guideline studies.

Here, we present a protocol for the isolation of whole, intact mouse mammary glands to investigate extracellular matrix (ECM) expression and ductal morphology.
