Dii Labeling

DiI labeling is a fluorescence-based method for marking cell membranes and tracing labeled cells over time, making it useful for studying cell behavior in cancer research. The lipophilic dye inserts into the plasma membrane, where its stable fluorescent signal can be detected by fluorescence microscopy or flow cytometry; as cells divide, the label is distributed between daughter cells and gradually diluted. Researchers use DiI-labeled tumor or stromal cells to monitor migration, invasion, proliferation, cell-cell interactions, and tissue distribution in culture or animal models. These measurements help clarify how cancer cells spread and respond to their microenvironment, supporting studies of metastasis and therapeutic strategies.

Here we report dual labeling of neural crest cells and blood vessels using chickGFP neural tube intraspecies grafting combined with intra-vascular DiI injection. This experimental technique allows us to simultaneously visualize and study development of the NCC-derived (enteric) nervous system and the vascular system, during organogenesis.

We present a method to visualize cuticle in live C. elegans using the red fluorescent lipophilic dye DiI (1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate), which is commonly used in C. elegans to visualize environmentally exposed neurons. With this optimized protocol, alae and annular cuticular structures are stained by DiI and observed using compound microscopy.
