Lyophilization Procedure

The lyophilization procedure, or freeze-drying, preserves biological materials and pharmaceutical formulations by removing water at low temperature while maintaining structural and functional stability. The process freezes the sample, lowers pressure to drive ice sublimation during primary drying, and then removes residual moisture through secondary drying. In cancer research, lyophilization supports the storage and transport of tissue-derived materials, proteins, antibodies, diagnostic reagents, and drug formulations that may be unstable in solution. By extending shelf life and improving consistency, this method helps maintain sample quality for molecular analysis, assay development, therapeutic evaluation, and reproducible experimental workflows.

Here, we present detailed protocols for solid-state amide hydrogen/deuterium exchange mass spectrometry (ssHDX-MS) and solid-state photolytic labeling mass spectrometry (ssPL-MS) for proteins in solid powders. The methods provide high-resolution information on protein conformation and interactions in the amorphous solid-state, which may be useful in formulation design.
