Microglia Conditioned Media

Microglia conditioned media is a cell-culture preparation containing soluble factors released by microglia into their surrounding medium, providing a controlled way to study microglia-mediated signaling. Researchers generate it by culturing microglia under defined conditions, collecting the medium, and applying it to other cells or experimental systems without transferring the microglia themselves. The medium can contain cytokines, chemokines, growth factors, and other secreted molecules that influence tumor cells, immune cells, endothelial cells, or neural cells. In cancer research, this approach helps characterize communication within the tumor microenvironment and assess how microglial activity may affect tumor growth, invasion, inflammation, and responses to treatment.

SEC-based Isolation of Microglia-derived Extracellular Vesicles: A Technique to Isolate Extracellular Vesicles from Microglia Conditioned Culture Media

2023

This video describes a method based on size-exclusion chromatography (SEC) to isolate extracellular vesicles or EVs derived from the brain immune cells―the microglia. The EVs act as molecular cargo to deliver mediators, which regulate an immune response. Therefore, the isolated EVs can be used as an in vitro model to study the microglia-neuron crosstalk during a neuronal immune response.

The protocol describes a reproducible method designed for use with cell culture supernatants to detect surface epitopes on small extracellular vesicles (EV). It utilizes specific EV immunoprecipitation using beads coupled with antibodies that recognize surface antigen CD9, CD63, and CD81. The method is optimized for downstream flow cytometry analysis.

Microglia (immune cells of the brain), are used as a surrogate biosensor to determine how nanoparticles influence neurotoxicity. We describe a series of experiments designed to assay microglial response to nanoparticles and exposure of hypothalamic neurons to supernatant from activated microglia to determine neurotoxicity.
