Organoid Formation Assay

An organoid formation assay is a three-dimensional culture method that measures how cells self-organize into organoids, providing a model of tissue structure and function in cancer research. Cells are embedded in a supportive extracellular matrix and maintained with defined growth factors, allowing stem-like or tumor cells to proliferate, differentiate, and form organized, often cystic or tissue-like structures. Researchers assess organoid number, size, morphology, and growth to evaluate tumor-initiating capacity, cellular heterogeneity, and responses to genetic or drug-based perturbations. Because organoids can retain features of patient tumors, this assay supports studies of cancer biology, therapeutic sensitivity, resistance mechanisms, and personalized treatment strategies.
