Targeted Dna Capture

Targeted DNA capture is a molecular technique that selectively enriches specific genomic regions from a complex DNA sample, enabling focused analysis of genes or variants relevant to cancer research. The method uses complementary oligonucleotide probes to hybridize with chosen DNA sequences, after which probe-bound fragments are isolated, commonly with biotin-streptavidin magnetic beads, and prepared for sequencing. By concentrating informative regions, targeted DNA capture improves sequencing efficiency and supports sensitive detection of mutations, copy-number changes, and other genomic alterations in tumor samples. This approach helps researchers characterize cancer genomes, compare tumor and normal DNA, investigate disease mechanisms, and evaluate potential biomarkers or therapeutic targets.

Primer Extension Capture: Targeted Sequence Retrieval from Heavily Degraded DNA Sources

We present a method of targeted ancient DNA sequence retrieval, which we used to reconstruct the complete mitochondrial genomes of five Neandertal individuals. Comparison of these sequences with present day humans suggests that Neandertals had a long term low effective population size.

Described is a two-step labeling process using β-glucosyltransferase (β-GT) to transfer an azide-glucose to 5-hmC, followed by click chemistry to transfer a biotin linker for easy and density-independent enrichment. This efficient and specific labeling method enables enrichment of 5-hmC with extremely low background and high-throughput epigenomic mapping via next-generation sequencing.
