Ammonium Formate Washing

Ammonium formate washing is a chemistry workup technique that uses an ammonium formate solution to rinse a reaction mixture, precipitate, or solid phase and remove soluble residues while retaining the desired material. As an ionic, water-compatible salt, ammonium formate helps dissolve and carry away unreacted reagents, by-products, and other contaminants; concentration, pH, and contact time influence washing efficiency and product recovery. This method can support purification when water or organic-solvent washes are insufficient, and its volatile character makes it useful in workflows that require low-residue sample preparation, including analytical mass spectrometry. Careful control of washing conditions improves separation and reproducibility.

Here, we describe a detailed protocol for the in vitro reassociation of purified, salt-washed eukaryotic ribosomal subunits for the analysis of 80S particle formation. This method is illustrated by analyzing ribosomal subunits isolated from a wild-type strain of Saccharomyces cerevisiae and a mutant strain deficient in the ribosomal protein eL24.

Washing, Drying, and Ignition of Precipitates

After filtration, the precipitate is washed to remove coprecipitated impurities and any remaining mother liquor. Colloidal precipitates, such as silver chloride, are washed with an electrolyte (such as dilute nitric acid) to prevent the peptization of the precipitate. In the case of slightly soluble precipitates, the wash solution contains a common ion to reduce solubility. Lead sulfate, which is slightly soluble in water, is washed with dilute sulfuric acid. Similarly, wash solutions may be...
