Caffeine Analysis

Caffeine analysis is the qualitative and quantitative determination of caffeine in materials such as coffee, tea, soft drinks, pharmaceuticals, and biological samples. In chemistry, analysts commonly isolate caffeine through solvent extraction and measure it using techniques such as ultraviolet-visible spectroscopy or high-performance liquid chromatography, where caffeine is identified by its characteristic signal and quantified against standards. Sample preparation, calibration, and separation help distinguish caffeine from other compounds in complex mixtures. These methods support product quality control, regulatory compliance, research on caffeine intake, and the development of reliable analytical procedures for consumer and pharmaceutical products.

This protocol describes an efficient methodology for the extraction and quantification of caffeine in cell suspensions of C. arabica L. and an experimental process for evaluating the enzymatic activity of caffeine synthase with the expression level of the gene that encodes this enzyme.

Experimental Protocol for Examining Behavioral Response Profiles in Larval Fish: Application to the Neuro-stimulant Caffeine

Here, we present a protocol to examine larval zebrafish and fathead minnow locomotor activities and photomotor responses (PMR) using an automated tracking software. When incorporated in common toxicology bioassays, analyses of these behaviors provide a diagnostic tool to examine chemical bioactivity. This protocol is described using caffeine, a model neurostimulant.

Analysis of Embryonic and Larval Zebrafish Skeletal Myofibers from Dissociated Preparations

Zebrafish are an emerging system for modeling human disorders of the skeletal muscle. We describe a fast and efficient method to isolate skeletal muscle myofibers from embryonic and larval zebrafish. This method yields a high-density myofiber preparation suitable for study of single skeletal muscle fiber morphology, protein subcellular localization, and muscle physiology.

A Neuronal and Astrocyte Co-Culture Assay for High Content Analysis of Neurotoxicity

This article describes a novel protocol and reagent set designed for sensitive measurement of neurotoxic effects of compounds and treatments on co-cultures of neurons and astrocytes using high content analysis. Results demonstrate that high content analysis represents an exciting novel technology for neurotoxicity assessment.

Automated Analysis of Intracellular Phenotypes of Salmonella Using ImageJ

Salmonella invades and replicates inside intestinal epithelial cells both in Salmonella-specific vacuoles and free in the cytosol (hyper-replication). A high-throughput fluorescence microscopy-based protocol is described here to quantify the intracellular phenotypes of Salmonella by two complementary image analyses through ImageJ, reaching single-cell resolution and scoring.
