Non-targeted Metabolite Profiling

Non-targeted metabolite profiling is an analytical strategy for surveying as many small-molecule metabolites as possible in a biological, environmental, or chemical sample without selecting targets in advance. The workflow typically combines sample extraction with liquid or gas chromatography and high-resolution mass spectrometry, or nuclear magnetic resonance spectroscopy, followed by signal processing, statistical comparison, and metabolite annotation. By capturing broad chemical features, the approach can reveal metabolic differences associated with disease, treatment, environmental exposure, or changes in experimental conditions. In chemistry research, it supports biomarker discovery, pathway analysis, quality assessment, and hypothesis generation, while complementary targeted methods are often used to confirm and quantify key findings.

We describe here the procedures for the extraction and purification of mRNA and metabolites from Drosophila heads. We are applying these techniques to better understand the cellular perturbations underlying neuronal degeneration. These methodologies can be easily scaled and adapted for other "omic" projects.
