Day 6 Differentiation

Day 6 differentiation is a defined time point in a developmental biology protocol when cultured cells begin or continue acquiring specialized identities from a less differentiated starting state. During this stage, coordinated changes in gene expression, cell signaling, morphology, and cell-cell interactions guide lineage commitment and help establish tissue-specific characteristics. Examining cells on day 6 allows researchers to monitor developmental progression, compare differentiation conditions, and assess the effects of growth factors or other experimental variables. This time point can support studies of embryonic development, disease modeling, regenerative biology, and the production of specialized cells for downstream analysis.

Differentiation of Human-Induced Pluripotent Stem Cells into Macrophages

This video describes an in vitro method for generating macrophages from human induced pluripotent stem cells (iPSCs). Macrophage generation from iPSCs occurs in three stages: (i) aggregation of iPSCs in suspension to form three-dimensional embryoid bodies (EBs) that differentiate into mesodermal cells, (ii) formation of macrophage precursor cells from the differentiated EBs, and (iii) differentiation of the macrophage precursor cells into mature, functional macrophages.
