Perfused Vasculature

Perfused vasculature is a functional network of blood vessels through which blood or a laboratory perfusate flows, delivering nutrients and oxygen while removing metabolic waste. Its function depends on interconnected, endothelial-lined channels that sustain fluid movement under a pressure gradient, enabling transport across vessel walls and exposing developing tissues to mechanical cues such as shear stress. In developmental biology, studying perfused vasculature helps reveal how vascular networks form, mature, and support organ growth, while perfusion models provide more physiologically relevant systems for investigating tissue development, vascular disease, and engineered tissues.

We show here the in vivo insertion of an osmotic pump for constant local drug delivery and the creation of hindlimb ischemia in a mouse model. Moreover, the hindlimb vasculature is perfused with Microfil, a silicone radiopaque agent, to prepare for micro-computed tomography (micro-CT) imaging.
