Lipid Coated Droplets

Lipid-coated droplets are liquid compartments surrounded by a self-assembled lipid layer, providing a controllable interface between aqueous and nonaqueous phases. At an oil-water boundary, amphiphilic lipids orient their hydrophilic heads toward water and hydrophobic tails toward oil, reducing interfacial tension and stabilizing droplets; microfluidic flow can further control droplet size and uniformity. In engineering, these structures enable compartmentalized biochemical reactions, artificial cell and membrane studies, high-throughput screening, and encapsulation of biological or chemical cargo. Their tunable composition and permeability also support the design of microscale reactors, sensing platforms, and emerging drug-delivery systems.

Lipid droplets are important organelles for the replication of several pathogens, including the Hepatitis C Virus (HCV). We describe a method to isolate lipid droplets for quantitative mass spectrometry of associated proteins; it can be used under a variety of conditions, such as virus infection, environmental stress, or drug treatment.

Lipid-Protein Membrane Structure-Function Characterization using Droplet Interface Bilayers

An experimental protocol is presented to assemble, electrically stimulate, and analyze gramicidin A-doped droplet interface bilayers. Lipid-protein structure-function relationships are quantified by measuring changes in membrane area, ionic flux, and single-channel conductance, and relating these responses to plasticity-like changes in ionic conduction in a membrane model inspired by electrical synapses.

Isolation of Cellular Lipid Droplets: Two Purification Techniques Starting from Yeast Cells and Human Placentas

Two techniques for isolating cellular lipid droplets from 1) yeast cells and 2) human placentas are presented. The centerpiece of both procedures is density gradient centrifugation, where the resulting floating layer containing the droplets can be readily visualized by eye, extracted, and quantified by Western Blot analysis for purity.
