Bisulfite Analysis

Bisulfite analysis is a molecular technique used to map DNA methylation, an epigenetic modification that can regulate gene activity without changing the DNA sequence. Treatment with sodium bisulfite converts unmethylated cytosines to uracils, which are read as thymines after PCR or sequencing, while methylated cytosines are comparatively protected and remain as cytosines. By comparing these sequence changes with the original DNA, researchers can identify methylated sites and estimate methylation levels across genes or genomic regions. In genetics, bisulfite analysis supports studies of development, genomic imprinting, disease-associated epigenetic changes, and the effects of environmental or cellular conditions on gene regulation.

DNA Methylation Analysis

Methylation at CpG dinucleotides is a chemical modification of DNA hypothesized to play important roles in regulating gene expression. In particular, the methylation of clusters of methylation sites, called “CpG islands”, near promoters and other gene regulatory elements may contribute to the stable silencing of genes, for example, during epigenetic processes such as genomic imprinting and X-chromosome inactivation. At the same time, aberrant CpG methylation has been shown to be associated with...

Here, we present a protocol to remove aldehydes and reactive ketones from mixtures by a liquid-liquid extraction protocol directly with saturated sodium bisulfite in a miscible solvent. This combined protocol is rapid and facile to perform. The aldehyde or ketone can be re-isolated by the basification of the aqueous layer.
