Elongation Complex

The elongation complex is a molecular assembly that extends a growing nucleic acid strand during gene expression, making it central to accurate genetic information transfer. In transcription, RNA polymerase forms the core of this complex and moves along the DNA template, adding complementary ribonucleotides to the nascent RNA while associated factors regulate pausing, processivity, and passage through chromatin. These coordinated interactions influence transcript length, gene activity, and the timing of downstream RNA processing. Studying elongation complexes helps researchers understand gene regulation, identify mechanisms underlying transcriptional defects, and evaluate potential targets for genetic and therapeutic research.

Here, we describe the assembly of RNA polymerase II (Pol II) elongation complexes requiring only short synthetic DNA and RNA oligonucleotides and purified Pol II. These complexes are useful for studying mechanisms underlying co-transcriptional processing of transcripts associated with the Pol II elongation complex.
