Nested Ligation-mediated Pcr

Nested ligation-mediated PCR is a targeted DNA amplification technique that combines adaptor ligation with two successive rounds of polymerase chain reaction (PCR) to detect or analyze specific genomic fragments. The method first ligates known DNA adaptors to restriction-enzyme-generated DNA ends, then uses adaptor-specific and locus-specific primers for an initial amplification; a second PCR with internal, or nested, primers increases target specificity and reduces nonspecific products. In genetics, this approach supports the analysis of genomic regions near known sequences, including mapping insertion sites, characterizing DNA rearrangements, and examining sequence variation when only limited genomic material is available.
