Biofilm Dispersion

Biofilm dispersion is the process by which microorganisms leave a surface-associated community and return to a free-living, or planktonic, state, making it a key transition in infection biology. Within a mature biofilm, environmental cues can activate enzymes and regulatory pathways that break down the extracellular polymeric substance matrix, reduce cell-cell attachment, and promote motility and cell release. This transition can seed new sites, alter exposure to immune defenses and antibiotics, and complicate chronic infections. Understanding biofilm dispersion helps researchers examine pathogen dissemination, identify molecular targets that limit spread, and design treatments that combine matrix-disrupting strategies with antimicrobial or immune-based therapies.

Detection of Biofilm Disassembly Through a Dispersion Assay

The video showcases a biofilm dispersion assay using a multi-well plate. Buffer and glucose treatment disassemble the biofilm, releasing bacteria, while bile salts induce stress, retaining bacteria in the protective extracellular polymeric substance. Higher dispersion with buffer and glucose confirms successful biofilm disassembly, estimated by colony forming units measurement.

Assessing Biofilm Dispersal in Murine Wounds

Here, we describe ex vivo and in vivo methods for assessing bacterial dispersal from a wound infection in mice. This protocol can be utilized to test the efficacy of topical antimicrobial and anti-biofilm therapies, or to assess the dispersal capacity of different bacterial strains or species.

Growth of Mycobacterium tuberculosis Biofilms

Mycobacterium tuberculosis forms drug tolerant biofilms when cultured in certain conditions. Here we describe methods for culturing M. tuberculosis biofilms and determining the frequency of drug tolerant persisters. These protocols will be useful for further studies into the mechanisms of drug tolerance in M. tuberculosis.
