Gfp Transgenic Parasites

GFP transgenic parasites are parasites genetically engineered to express green fluorescent protein (GFP), providing a visible marker for studying their biology and interactions with hosts. Researchers introduce a GFP-encoding gene into the parasite genome or an expression system, allowing infected cells or organisms to fluoresce when the protein is produced under suitable conditions. In immunology and infection research, this labeling supports visualization and tracking of parasite development, tissue localization, transmission, and clearance by the host immune response. GFP transgenic parasites also enable quantitative assays of infection and evaluation of antiparasitic treatments, linking microscopic observations with disease mechanisms and therapeutic outcomes.

In vivo Imaging of Transgenic Leishmania Parasites in a Live Host

An in vivo imaging system is used to generate quantitative measurements of murine infection with the Trypanosomatid protozoan Leishmania. This is a non-invasive and non-lethal method for detecting parasites expressing luciferase within many tissues throughout the course of chronic Leishmania spp. infection.

We established the photoconvertible PSmOrange system as a powerful, straight-forward and cost inexpensive tool for in vivo cell tracking in GFP transgenic backgrounds. This protocol describes its application in the zebrafish model system.
