Hemagglutinin Expression Transfection

Hemagglutinin expression transfection is a laboratory method used to introduce a hemagglutinin-encoding nucleic acid into cultured cells so they produce this viral surface glycoprotein. After delivery into the cells, the genetic construct is transcribed and translated, directing hemagglutinin synthesis and, when appropriate, its transport to the cell membrane. Because hemagglutinin mediates viral attachment and membrane fusion, expressed protein provides a controllable system for examining viral entry, antigen recognition, and host-cell interactions. In immunology and infection research, the method supports antibody characterization, vaccine-related studies, and analysis of hemagglutinin function without requiring propagation of complete infectious virus.

Here we describe a way to express correctly folded and functional influenza virus surface antigens derived from the novel Chinese H7N9 virus in insect cells. The technique can be adapted to express ectodomains of any viral or cellular surface proteins.

In this manuscript, we present a time efficient method to achieve tightly controlled ion channel expression by performing transient transfection under the control of an inducible promoter.

We describe a method to measure the activation of Fc-mediated effector functions by antibodies that target the influenza virus hemagglutinin. This assay can also be adapted to assess the ability of monoclonal antibodies or polyclonal sera targeting other viral surface glycoproteins to induce Fc-mediated immunity.
