Human Enteroid Model

A human enteroid model is a three-dimensional, laboratory-grown representation of the human intestinal epithelium that reproduces key features of gut structure and function. It is typically generated from human intestinal stem cells, which proliferate and self-organize in a supportive culture environment into polarized epithelial tissues containing multiple intestinal cell types. These models provide a controlled system for examining epithelial barrier function, host-microbe interactions, pathogen invasion, and immune signaling without relying solely on animal models. In immunology and infection research, human enteroids help clarify disease mechanisms, evaluate antimicrobial or anti-inflammatory interventions, and support studies of personalized intestinal responses.

Enteroids are emerging as a novel model in the study of human disease. The protocol describes how to simulate an enteroid model of human necrotizing enterocolitis using lipopolysaccharide (LPS) treatment of enteroids generated from neonatal tissue. Collected enteroids demonstrate inflammatory changes akin to those seen in human necrotizing enterocolitis.

We describe a method to establish human enteroids from small intestinal crypts and colonoids from colon crypts collected from both surgical tissue and biopsies. In this methodological article, we present the culture modalities that are essential for the successful growth and maintenance of human enteroids and colonoids.

This protocol describes an in vitro model of necrotizing enterocolitis (NEC), which can be used for mechanistic studies into disease pathogenesis. It features a microfluidic chip seeded with intestinal enteroids derived from the human neonatal intestine, endothelial cells, and the intestinal microbiome of a neonate with severe NEC.

Here, we present a systematic approach for developing physiologically relevant, sensitive and specific in vivo assays for interpreting variation in human pathology. Transient genetic manipulation via microinjection of WT and mutant human mRNA and morpholino (MO) antisense oligonucleotides harness the tractability of the developing zebrafish embryo to rapidly assay pathogenic mutations, especially, but not exclusively, in the context of human developmental disorders.
