Interferon Subtype Expression

Interferon subtype expression is the regulated production of distinct interferon proteins by cells, shaping antiviral and immune responses during infection. Pathogen-associated molecular patterns activate sensors such as RIG-I-like receptors and Toll-like receptors, which induce transcription of interferon genes; the resulting subtypes bind specific receptors and trigger JAK-STAT signaling to regulate interferon-stimulated genes. Comparing subtype expression across infected tissues, cell types, and disease stages helps clarify how immune protection is coordinated and why responses differ among pathogens. These analyses support infection research, biomarker development, and evaluation of interferon-based therapies while informing strategies to balance antiviral activity with inflammatory tissue damage.

High-throughput Quantitative Real-time RT-PCR Assay for Determining Expression Profiles of Types I and III Interferon Subtypes

This protocol describes a high-throughput qRT-PCR assay for the analysis of type I and III IFN expression signatures. The assay discriminates single base pair differences between the highly similar transcripts of these genes. Through batch assembly and robotic pipetting, the assays are consistent and reproducible.

Expression Analysis of Mammalian Linker-histone Subtypes

We describe a set of assays to analyze expression levels of H1 linker histones. mRNA of individual H1 genes are quantitatively measured by random primer based reverse transcription followed by real-time PCR, whereas protein quantification of H1 histones is achieved by HPLC analysis.
