Murine Lung Imaging

Murine lung imaging is the visualization and measurement of lung structure, function, and biological activity in mice, providing a window into immune responses and respiratory infection. Depending on the modality, images capture anatomical changes or signals from fluorescently or bioluminescently labeled cells, microbes, or molecular probes in living animals or excised tissue. These approaches can track pathogen localization, leukocyte recruitment, inflammation, and tissue damage over time, linking spatial observations with disease progression. By enabling longitudinal analysis in the same animal, murine lung imaging supports infection modeling, evaluation of therapies, and refinement of experimental designs while reducing reliance on endpoint measurements.

We describe a relatively simple method for ex vivo live imaging of the tumor cell-stroma interactions within lung metastasis, utilizing fluorescent reporters in mice. Using spinning-disk confocal microscopy, this technique enables visualization of live cells for at least 4 hr and could be adapted to study other inflammatory lung conditions.
