Vascular Perfusion Fixation

Vascular perfusion fixation is a tissue-preservation technique that delivers a chemical fixative through the vascular system, maintaining anatomical structure and cellular detail throughout an organism or organ. After blood is displaced, controlled pressure drives the fixative through arteries, capillaries, and veins, where agents such as aldehydes cross-link proteins and stabilize tissues; fixation conditions must be optimized because excessive cross-linking can reduce antigen accessibility. In immunology and infection research, this approach supports reproducible histology, immunostaining, and analysis of pathogen distribution, inflammatory infiltrates, and host tissue responses while minimizing postmortem degradation and preserving spatial relationships.

Air-Inflation of Murine Lungs with Vascular Perfusion-Fixation

Presented is a method for air-inflation with vascular perfusion-fixation of the lungs that preserves the location of cells within airways, alveoli and interstitium for structure-function analyses. Constant airway pressure is maintained with an air-inflation chamber while fixative is perfused via the right ventricle. Lungs are processed for histologic studies.

The porcine model of liver normothermic machine perfusion (NMP), described here, can be successfully used to study NMP as a preservation strategy, a tool for viability assessment, and a platform for organ repair.
