Pathogen Strain Variants

Pathogen strain variants are genetically distinct forms of the same infectious organism that can differ in transmissibility, disease severity, immune recognition, or response to treatment. They arise through mechanisms such as mutation, genetic recombination, and natural selection, which can alter surface antigens, replication efficiency, or other traits during transmission and infection. In immunology and infection research, characterizing strain variants helps explain why immunity may vary between individuals or pathogens, supports molecular surveillance, and informs vaccine and therapeutic development. Comparing variants also reveals how pathogens adapt to host defenses and changing environmental pressures, strengthening outbreak response and infectious disease control.

This video demonstrates a technique for generating escape variants of neutralizing influenza virus monoclonal antibodies. Antibody-neutralized virus suspension is injected into embryonated chicken eggs to propagate escape variants selectively. This results in a virus population predominantly of escape variants, confirmed via a Hemagglutination assay.

Single Cell Measurements of Vacuolar Rupture Caused by Intracellular Pathogens

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.

We describe a method by which we identify critical residues required for the binding of human or murine monoclonal antibodies that target the viral hemagglutinin of influenza A viruses. The protocol can be adapted to other virus surface glycoproteins and their corresponding neutralizing antibodies.
