Permeable Supports

Permeable supports are porous membranes or scaffolds that enable controlled exchange between separated biological compartments, making them useful for modeling barriers and transport in immunology and infection research. Their interconnected pores permit soluble factors, nutrients, and metabolites to diffuse while retaining cells or microorganisms according to pore size and experimental design. In culture systems, these supports can separate immune and target cells, allowing researchers to assess chemokine-driven migration, barrier permeability, inflammatory signaling, and pathogen passage under defined conditions. By reproducing aspects of tissue interfaces, permeable supports help clarify host-pathogen interactions and evaluate immune responses, antimicrobial treatments, and strategies for strengthening biological barriers.

Here, we present a protocol describing the culture of canine intestinal organoids in a dual-chamber, permeable support system. Organoid seeding in the permeable supports, the monolayer maintenance, and subsequent drug permeability experiments are described.

An Assay for Permeability of the Zebrafish Embryonic Neuroepithelium

We describe a live whole animal quantitative measurement for permeability of the embryonic zebrafish brain. The technique analyzes the ability to retain cerebrospinal fluid and molecules of different molecular weights within the neural tube lumen and quantifies their movement out of the ventricles. This method is useful for determining differences in epithelial permeability and maturation during development and disease.
