Plasmacytoid Dendritic Cells

Plasmacytoid dendritic cells (pDCs) are specialized innate immune cells that detect nucleic acids and rapidly coordinate antiviral defenses. In response to viral RNA or DNA, endosomal Toll-like receptors 7 and 9 activate signaling pathways that trigger the production of large amounts of type I interferons, particularly interferon-alpha. These cells also present antigens and interact with natural killer cells, T cells, and other immune populations to shape adaptive immunity. Studying pDC biology helps clarify host responses to viral infection and informs research on autoimmune disease, cancer immunology, vaccine development, and therapies that modulate interferon-driven inflammation.

In Vitro Generation of Plasmacytoid Dendritic Cells from Common Lymphoid Progenitors

This video demonstrates an in vitro assay to generate plasmacytoid dendritic cells (pDCs) from common lymphoid progenitor cells (CLPs). Upon co-culturing purified CLPs with γ-irradiated feeder cells in the presence of Fms-like tyrosine kinase-3 ligand (FL), the CLPs differentiate into non-adherent pDCs, the presence of which is confirmed using flow cytometry.

Unlike cell-free HIV-1 particles, infected CD4+ T cells are effectively sensed by plasmacytoid dendritic cells (pDCs). This manuscript describes a method where peripheral blood mononuclear cells (PBMCs) or isolated pDCs are co-cultured with HIV-1 infected T cells to evaluate innate sensing by pDCs as assessed by release of type-I IFN.
